To begin in a very general sense, Deus Fortress is a multiplayer mod that combines the world of Deus Ex with the gameplay of Team Fortress. For those of you unfamiliar with what Team Fortress is, in a nutshell it provides team-based play centered around player 'classes', where each class has it's own unique and very separate abilities. Working as a team is vital in order to best capitalize on everyone's proficiencies.
So, what about DXF?
The guys gave us a little bit of background:

[PDX]How did Deus Fortress come into existence?
[G] You'll have to ask EternalSpark, since this was his baby. I can tell you how I came to join the team, though -- I saw an interview that mentioned Deus Ex, my favorite game, and Team Fortress, quite possibly the best "classic" PC game since Wasteland, in the same sentence, and immediately emailed the author. I got EternalSpark's email address, and after a few messages shot back and forth, I was in.
[EternalSpark] 'The "birth thought", so to speak, was after I played Deus Ex for about the 3rd time. I made a joke to a friend of mine, something to the extent of "heh, I played as a Heavy Weapons guy that time!" - that got me looking at the skills in the game, and the thought struck me - not only could I make a TF mod which has all the classes which TF fans know and love - [I could make] new ones as well. Instantly I was thinking about how to do a DX TF mod. Not one that is a mere rehash of current TF, or TF mods with one or 2 new things but the rest being the old vanilla - but something WAY new. DX itself broke so many barriers we thought games would never get to - and I couldnt help think that this could really do some good for the community.
So, I posted a message on Deus-ex.org's forums, basically asking "hey guys, I think a new TF /DX mod would be cool, what do you think?". The rest, as they say, was history. :)'
The Five Game Modes
I've heard the Deus Fortress guys say time and time again that they aren't trying to just rehash or copy existing TF design to their project. Let's take a look at what the mod will actually contain, and then you can decide if this is just "TF with Sunglasses".
First off, the gametypes. Naturally they have an ambitious project scope, so this incorporates several different modes of play.
Blood of the Savior Two teams, two types of fluid (from the blood of of HelioDenton). Help your team collect the fluid you need, while stopping your opponents from getting theirs.
Grey Death Get a cannister of virus, flood the entire area, then find the single cannister of antidote and get it back to your base to stay alive.
Area Conquest Find the few remaining computers and program them, while deprogramming any that your opponents got to first.
Lifeblood War Keep your UC active, destroy your opponents, and then wipe them out. Get the UC first or they'll just keep regenerating.
Covert Both teams are trying to evacuate their key team Nanomancer: Save yours, kill theirs.
Whew! Not your usual circle-strafe fragfest, and it really seems like the guys are keepin' it real and putting their own unique touch on the general TF idea.
Classes Galore
None of this would be fun without all the cool and differentiated player types, though. The nice new twist is that, in addition to player classes differentiated by skill settings, players will also be able to choose from several different augs. This will effectively allow creating class subsets, making it impossible to know just what you're facing off against by look alone.
Not much is public at this point about the augmentation system, but Deus Fortress has put up a detailed Class breakdown page, complete with images and everything you need to know. You'll get to choose from the following list.
